Scope of work From 2010 to 2013, Multiconsult was responsible for the detailed design of the upgrading of civil structures, the optimised design of steel and civil works, hydraulic modelling, updating of the hydrology of Lake Malawi/Shire River basin, power simulations, detailed design of hydrological measurements stations and data transmission system for operation of the Kamuzu Barrage; Environmental Impact Assessment, economic analysis and preparation of model tool for hydrological forecasting and optimized regulation (KABOM – Kamuzu Barrage Operation Model), as well as preparation of bid documents. From 2014, Multiconsult has been responsible for the supervision and contract management of all construction works as well as supply and installation of new gates, new weed boom and weed collection arrangements; and supervise Environmental, Health and Safety issues on site and in the local community. The development objectives of Sindh Barrages Improvement Project for Pakistan are to improve the reliability and safety of the Guddu barrage and strengthen the Sindh Irrigation Department’s capacity to operate and manage the barrage. This project requires additional financing (AF) to scale up activities under the original project. New activities under the this AF will include (a) rehabilitation and modernization of Sukkur Barrage; (b) support for the integrated Operation and Maintenance (O and M) of the three barrages in Sindh Province; (c) critical studies to improve coordination and management of the three barrages; and (d) better management of the riverine habitats of the main Indus between Guddu and Sukkur Barrages. The project will have a closing date of June 30, 2024. The project components will be restructured as follows, including two new components. 1) The first component, Rehabilitation of Guddu and Sukkur Barrages, would support the rehabilitation of Sukkur Barrage as well as Guddu Barrage. 2) The second component, Improved Barrage Operation, aims to provide technical assistance and equipment support to facilitate the O and M of Guddu Barrage. 3) The third component, Technical Studies, aims to help deepen GoSindh’s technical understanding of barrages and prepare for Phase 2. 4) The fourth component, River Basin Water Resources and Riverine Management, aims to sharpen the focus on the water-related environmental and social issues in the 170-kilometers stretch of the Indus between Sukkur and Guddu Barrages, which also is a designated Ramsar site. 5) The fifth component, Project Management, Monitoring, and Evaluation, aims to support Sukkur Barrage rehabilitation. Living cells are constantly barraged by perturbations that originate within themselves. Especially abundant – far more than DNA mutations – are two kinds of such perturbations. The first is gene expression noise, pervasive stochastic variation of transcript and protein levels. The second is mistranslation noise, the misincorporation of amino acids by ribosomes during protein synthesis. Organisms and protein molecules can evolve robustness – the persistence of well-adapted phenotypes – to both kinds of noise. Theory predicts that noise and robustness can affect the adaptive evolution of new proteins, but we do not know whether they help or hinder adaptive evolution. We hypothesize that noise and robustness can accelerate protein evolution both separately and jointly. To validate this hypothesis, we will evolve light-emitting fluorescent proteins towards new color phenotypes via directed laboratory evolution in E.coli. During evolution, we will manipulate expression noise by driving FP expression from noisy or quiet promoters, and we will manipulate mistranslation via host strains with low and high mistranslation rates. We will manipulate protein robustness in three biologically important ways, chaperone overexpression, gene duplication, and stabilizing selection. We will study how fast FPs evolve new colors, and analyze protein evolutionary dynamics through a combination of high-throughput sequencing, engineering of selected adaptive mutations, and data-driven modeling. Our project will show how a ubiquitous but poorly understood source of phenotypic variation affects protein innovation. It will also help engineers discover new protein functions. Moreover, our work will help establish FPs as a major platform to study protein evolutionary dynamics. By revealing noise as a new and crucial factor in protein evolution, our observations have the potential to revolutionize molecular evolution research, much like earlier studies of noise have revolutionized cell biology.

There are three components to the project, the first component being rehabilitation of the Guddu barrage. This component will support rehabilitation of the barrage and its associated structures. The component will finance: concrete repairs along the upstream gate grooves and rehabilitation of both fish ladders; removal of the left pocket dividing wall and construction of a new left pocket dividing wall to widen the left pocket and improve its ability to reduce the quantities of sediment entering the Ghotki feeder canal; and river training works through construction of a left-bank spur to promote favorable river approach conditions to the barrage and strengthen the upstream existing river training works. The second component is the improved barrage operation. This component will support the establishment of the BMO and modernization and improvements to the barrage O&M. Institutionally this will include undertaking a study to design the BMO structure, defining roles and responsibilities, and moving towards a more service oriented culture. Technically the Bank will finance necessary upgrades to the monitoring instrumentation, such as piezometers, gate positioning and water measurement equipment, replacement of surveillance and maintenance boats, and procurement of hydrographic equipment. The project will support workshops and provide a stock of spare parts, for use in maintenance activities. The instrument monitoring system for the barrage will be renovated, and the operations staff will be equipped with an updated operation, maintenance, and surveillance manual. The objectives of the Second Phase of the Punjab Barrages Improvement Project for Pakistan are to assist the Borrower in: (i) rehabilitating and modernizing Jinnah Barrage and carrying out affiliated works to enable reliable and uninterrupted supply of water for over 2.1 million acres of farmland benefitting about 600,000 farm families for irrigation and domestic water users; and (ii) build Irrigation and Power Department (IPD) capacity in improved water resources and irrigation system management. There are four components to the project, the first component being rehabilitation and modernization of Jinnah barrage. This component will support rehabilitation and modernization of Jinnah barrage, the implementation of social and environmental management plans, and construction supervision and support for the project s preparation and implementation. The Project Management Organization (PMO) of the Punjab s Irrigation and Power Department (IPD) will be responsible for implementation of this component. The second component is the improvement and modernization of the irrigation and water management system.
